Yes, the primary loop pump would pump water to the tank and then to the primary loop again where the pump used to attach; in other words, the circulator supply is disconnected, connected to the tank and the tank is connected to the primary piping where the pump supply was attached.

The boiler inputs 240,000 btuh or 4000 btu per min. An 80 gal tank would have 80 x 8.33 lbs of water times 20 degrees = 13,328 btuh to heat up which gives the boiler 3 minutes to run and temper. Actually, if the tank is up to temperature and the house is working on outdoor reset, the system can be set up to use the tank for heat for 3 minutes and draw the tank down in temperature before the burner has to kick on. This could mean that water draws by the indirect Phase 3 could be partially heated before the burner needs to go on. There are advantages. There isn't much more gained by adding a minute with a larger 100 gallon tank.
