Check the pressure on the second floor bleeder; water should spray out quickly. If water just dribbles out, increase the pressure in the system. To see if there is an air block in the lines, increase the pressure in the system to 20 psig on the gauge on the boiler, the air might then move out when it is squeezed to a smaller volume wherever it is and the upstairs loop run normally.
